TRIzol LS Reagent is suitable for extraction of total RNA from various types of liquid samples, with a wide range of applicability. Samples can originate from humans, animals, plants, yeast, bacteria, viruses, serum, plasma, whole blood, body fluids, etc., and it yields good extraction integrity. The final volume ratio of TRIzol LS to liquid sample is 3:1. It provides effective separation for small amounts of tissue (50–100 mg) and 5×10^6 cells, as well as for large amounts of tissue (≥1 g) and cells (>10^7).
While samples are thoroughly lysed inTRIzol LS Reagent, RNA integrity is maximally preserved. After addition of chloroform and centrifugation, the solution separates into three phases: an upper colorless aqueous phase, an interphase, and a lower organic phase, with RNA located in the upper aqueous phase. After collecting the aqueous phase, total RNA can be recovered by isopropanol precipitation.
Applications
The extracted totalRNA has good integrity, with no protein or DNA contamination, and can be used for RT-PCR, Real-time RT-PCR, Northern blot, Dot Blot, in vitro translation and other molecular biology experiments.
